西伯利亚蓼 PsGRX 基因的克隆及其在 NaHCO3 胁迫下的表达

摘 要:

采用 cDNA 末端快速扩增(RACE)技术从西伯利亚蓼叶 cDNA 文库中克隆到谷氧还蛋白基因(PsGRX)的完整编码区 cDNA 序列(GenBank 注册号为 GU139794), 长度为 465 bp, 编码 106 个氨基酸。根据与其他植物谷氧还蛋白的氨基酸序列 的比对以及系统进化分析的结果, 初步确定此基因为谷氧还蛋白基因家族成员。实时定量PCR的结果显示, PsGRX在西伯 利亚蓼的叶、茎、地下茎中均有表达, 叶中表达量最高, 地下茎和茎中较低。在 NaHCO3 胁迫的过程中, 此基因在叶、茎和 地下茎中的表达模式也有较明显的差异。

Cloning and Analysis of PsGRX Gene from Polygonum sibiricum Laxm. and Its Expression in Response to NaHCO3

Abstract:

The PsGRX gene (GenBank accession No. GU139794) which contained a complete open reading frame (ORF) was obtained using rapid amplification of cDNA ends (RACE) from the cDNA library of Polygonum sibiricum leaves. The length of cDNA was 465 bp, encoded a peptide of 106 amino acid residues. Based on the comparison with amino acid sequences of other plant glutaredoxin and the phylogenetic analysis of protein evolution, this gene was divided into glutaredoxin family. Expression analysis with real-time PCR showed that PsGRX gene was expressed in leaves, stems and underground stems. The expression level of PsGRX gene was higher in leaves than in stems and underground stems. The expression patterns of PsGRX gene in different tissues induced by NaHCO3 stress also had remarkable differences.
